Abstract:

PROBLEM TO BE SOLVED: To readily obtain a train of chromosome DNA having a centromere region, a telomere region and a replication-initiating region by polymerizing deoxyribonucleotide in a reaction system in the absence of a template and a primer, etc., and in the presence of a protein.
SOLUTION: In a reaction system not containing a template and/or a primer, a manifesting vector containing a DNA coding a protein composed of a heat- resistant deoxyribonucleotide polymerase, etc., of a bacterium belonging to genera Thermococcus, Thermus or Pyrococcus is introduced into a cell and the protein is produced in the cell, then the deoxyribonucleotide is polymerized in the presence of the protein at 60-90°C to readily obtain the objective chromosome DNA composed of a train of double stranded linear DNA having a centromere region, a telomere region and a replication-initiating region as necessary elements in the chromosome.
