Abstract:
Lyophilized restriction endonuclease is new. The lyophilized restriction endonuclease is produced by (a) preparing a mixture of a restriction endonuclease, a lyophilizable solubiliser and a freezable liq., (b) rapidly freezing the mixt., and (c) dehydrating the frozen mixt. The freezing and dehydration are pref. performed practically simultaneously, and the final moisture content of the hyophilizate is pref. ca 4wt.%. Provided appropriate conditions are used, restriction endonucleases can be lyophilized (freeze-dried) without loss of enzymatic activity to give lyophilizates which are not subject to the instability (attributed to contamination by proteases) of the corresponding liq. preparations and which therefore do not need to be kept at low temp.
