Abstract:

To provide a heat-resistant DNA ligase improved in bindability to DNA and in reaction activity.

The modified heat-resistant DNA ligase is obtained by replacing a negatively charged amino acid (for example, an amino acid corresponding to the 540-aspartic acid of the sequence number 1) existing on the N-terminal end side of the C-terminal helix portion of a heat-resistant DNA ligase originated from thermophilic bacterium, ultrathermophilic bacterium, thermophilic archaebacterium, or ultrathermophilic archaebacterium by a negative charge-free amino acid (for example, alanine, serine, arginine or lysine), and has more improved DNA bindability than those of wild type bacteria.
