Abstract:

PURPOSE: To provide a protein saccharification rate measuring method without requiring independent measurement of a total protein quantity, which is applicable to general purpose, inexpensive, simple and excellent in reproducibility, calibration curve linearlity and the like.
CONSTITUTION: Aqueous solutions of 4g/dl of unsaccharified human hemoglobin and nonenzymatic saccharified human hemoglobin are mixed with each other to prepare samples wherein rates of the saccharified human hemoglobin are 12, 25, 35%. Respective samples are dispensed into holes of microplates (solid phase) to physically adsorb the human hemoglobin. After cleaning by physiological saline, 1% beef blood albumin aqueous solution is dispensed to physically adsorb the beef blood albumin. After cleaning with the physiological saline, solution of peroxidase labelled phenylboronic acid derivative is added, cleaning is conducted after reaction, o-phenylenediamine solution is added. After reaction, the reaction is stopped by sulfuric acid and absorbance at measurement wave length 492nm is measured.
