Abstract:
PURPOSE: To obtain a sufficiently defined peptide profile with good reproducibility by mixing a protein hydrolase with a protein substrate, performing a first hydrolysis process in an agitation tank and also performing a second hydrolysis process within a tube provided with a stationary mixing element.
CONSTITUTION: This method comprises: supplying measured amounts of a protein substrate and a protein hydrolase to an agitation tank 1 from a vessel 7 and a vessel 13 respectively to perform a first hydrolysis process there while agitating them with an agitator 3; and then, transferring the first hydrolysis product to a tube 25 provided with a stationary mixing element 27 to perform a second hydrolysis process there. At this time, the residence time of the total amount of the substrate can accurately and simply be adjusted.
