Abstract:

To provide a method of measuring a sugar chain with high sensitivity using a mass analyzer for detecting a very small amount of the sugar chain.

A method for the high sensitivity MALDI mass analysis of the sugar chain includes: the step (i) for forming the liquid droplet of a mixed solution, wherein a sample to be analyzed containing a sugar chain molecule, a liquid matrix being a liquid substance containing aminoquinoline and an acidic group-containing substance and an ammonium salt being an additive are contained in a solvent, on a target plate; the step (ii) for removing the solvent from the liquid droplet of the mixed solution to contract the contact area of the target plate and the liquid droplet of the mixed solution along with the reduction of the volume of the liquid droplet to thereby gather the sample to be analyzed, ionic liquid and additive contained in the mixed solution to a part of the contact area and obtaining the focus spot of the mixed solution of the sample to be analyzed, the ionic liquid and the additive; and the step (iii) for subjecting the focus spot of the mixed solution to MALDI mass analysis measurement.
