PURPOSE: To sufficiently extend the life of excited state for multiple resonance absorption even under weak lighting and increase the absolute number of excited molecules by dipping a biological specimen in solution having a pH value and temperature in specific ranges.
CONSTITUTION: A biological specimen is dipped in solution having a pH value in about 0.5 to 10 and temperature of about 5°C or less. By this, the life of excited state can be extended sufficiently long for multiple resonance absorption even with weak luminous intensity of lighting. Therefore, sufficient excited molecules are obtained even under weak light and adverse effect to a biological specimen which was unavoidable under high luminous intensity lighting, is eliminated. Also, large sizing of light source is prevented and the life of excited state is prolonged and so an image with good contrast and exactness is obtained.
