Abstract:

PROBLEM TO BE SOLVED: To obtain a new compound having an excellent antibacterial activity against Gram negative and a Gram positive bacteria, also being extremely low toxic, having an excellent peroral absorptivity, and useful as a synthetic antibacterial agent.
SOLUTION: This new pyridone carboxylic acid derivative is a compound of formula I [X is N; Y is CH, CR7 (R7 is a lower alkyl or a halogen); Z is CH; R2a is a (substituted) amino; R3 is H, a halogen], e.g. ethyl 1-[6-(t- butylamino)-3,5-difluoropyridin-2yl]-8-chloro-6,7-difluoro-4-oxo-1,4- dihydroquinoline-3-carboxylate, etc. Further, in order to obtain the above compound, it is preferable pass through an acrylic acid ester derivative obtained by reacting a compound of formula II [R1a is a lower alkyl; L1, R5a are each a halogen; W is N-atom or CR8 (R8 is H, a halogen or the like)] with orthofornic acid.
