Abstract:

PROBLEM TO BE SOLVED: To obtain an α-haloacetophenone derivative useful as an intermediate for a medicine, etc., by a simple and safe process in high yield and selectively.
SOLUTION: A methylthioacetophenone of formula I (X is Cl or Br) as a raw material is halogenated with Cl or Br to give an α-halo(methylthio) acetophenone of formula II. In the operation, the amount of the alcohol is generally 1-20 times as much as the compound of formula I, that of the halogenating agent is 0.8-2 times mol of the compound. The reaction is carried out at -20 to 120°C. Then, the compound of formula II is reacted usually with 1.5-10mol equivalent of an aqueous solution of H2O2 as an oxidizing agent in the presence of Na2WO4 in an amount of 0.001-0.1 as much as the α-halo(methylthio) acetophenone further by using an acid such as hydrochloric acid at about 0-100°C for about 1-10 hours to give the objective α-halo(methylsulfonyl) of formula III. The compound of formula III is isolated by extraction, crystallization, etc.
