Abstract:

PURPOSE: To obtain a vector to secrete a formed protein to the outside of a cell, by inserting a gene to code a precursor protein wherein a C end amino acid of OmpF signal peptide is directly bonded to an N end amino acid of desired protein into the downstream of a promoter.

CONSTITUTION: A signal peptide gene of OmpF such as psp64OmpF No.21 deficient in PstI site of the upper stream of OmpF promoter, etc. is directly bonded to a structural gene of β-endorphin to code desired protein. For example, smooth end-SalI end DNA fragment of 138bp is obtained from plasmid pYT3 containing a structural gene of β-endorphin, which is linked to a vector fragment consisting of smooth end-SalI end obtained from plasmid psp640mpF No.21 with T4DNA ligase, and transformed into strain RRI of Escherichia coli to give plasmid pNM200.
