Abstract:
Disclosed are a constant-temperature amplimer for nucleic acids and the use thereof. The amplimer comprises a forward primer and a reverse primer used for amplifying a target gene. The forward primer is a sequence having an exogenous self-paired hairpin structure at a 5' terminal, or a sequence having, at the 5' terminal, a hairpin structure formed by matching with any source sequence located in the target gene and close to a forward primer terminal. The present invention has the effects as follows: the amplimer of the present invention is used for producing the sequence tandem repeat structure of the target gene by means of nucleic acid amplification technology, and a repetition frequency can be precisely controlled; the technology also can be used in three-generation sequencing technology; and amplification steps are simple and fast, and the accuracy of sequencing is improved.
