Abstract:
Provided are a construction method for and an application of an engineered strain of Escherichia coli that fixes CO2 and produces malic acid, which belong to the field of fermentation. The engineered bacteria is bacteria obtained by performing genetic engineering modification on Escherichia coli MG1655. Said genetic engineering modification comprises knocking out a fumaric acid reductase gene, a fumarase gene, a lactate dehydrogenase gene and an alcohol dehydrogenase gene, and overexpressing formate dehydrogenase, acetyl-CoA synthase, acylated acetaldehyde dehydrogenase, formaldehyde lyase, dihydroxyacetone kinase, malic enzyme and phosphorous oxidoreductase so as to obtain a strain GH0407. The described strain is used for the fermentation production of malic acid. By using CO2 and glucose as co-substrates, anaerobic fermentation is carried out for 72 hours. The output of malic acid can reach 39g/L, and the yield is 1.53 mol/mol, whereas the original starting strain does not accumulate malic acid.
