Abstract:
Provided is immunochromatography having a high detection sensitivity. The immunochromatography involves: a mixing step of mixing a sample that may contain an antigen with modified magnetic particles which are magnetic particles modified with a substance having an affinity specifically for the antigen, thereby obtaining a mixture containing a magnetic particle complex; a collection step of using magnetism to collect the magnetic particles; a separation step of mixing the collected magnetic particles with a separation solution in an amount that is less than the sample that may contain the antigen to separate the modified magnetic particles and obtain an antigen concentrate solution; a neutralization step of obtaining a neutralized antigen concentrate solution; a development step of forming a gold particle complex, which is a complex between the antigen in the neutralized antigen concentrate solution and modified gold particles that are gold particles modified with a first binding substance, and in this state, proceeding with development into an insoluble support having a reactive site wherein a second binding substance has been fixed; a trapping step of trapping the gold particle complex in the reactive site; and a silver enhancement step of enhancing the gold particle complex with silver.
