Abstract:
Disclosed are a method for analyzing an oligonucleotide sequence impurity based on high throughput sequencing and the use. The method comprises the following steps: constructing a high throughput sequencing library for analyzing an oligonucleotide sequence impurity; performing high throughput sequencing on the high throughput sequencing library, and analyzing the components of the oligonucleotide sequence according to the sequencing results; the extended primer sequences used for constructing the high throughput sequencing library successively consists of DNA molecules shown in position 1-22 of Sequence 2 and N of base A or base T or base C or base G; and N is an integer greater than or equal to 6. Experiments demonstrate that the method for analyzing an oligonucleotide sequence impurity based on high throughput sequencing can quickly, accurately and completely analyze the purity and content of each component in the oligonucleotide sequence.
