The present invention addresses the problem of providing a method for introducing a substance into a protoplast or an alga by electroporation under novel voltage applying conditions.　The problem can be solved by a method for introducing a substance into a protoplast or an alga by electroporation, the method comprising a mixed liquid placement step of placing a mixed liquid containing a protoplast or an alga and a substance to be introduced between a pair of electrodes for electroporation and a voltage application step of applying a voltage between the pair of electrodes for electroporation, wherein the voltage application step comprises one or more poration pulse application steps of applying a poration pulse of a negative voltage following a poration pulse of a positive voltage to one of the pair of electrodes for electroporation, and the interval from the application of the poration pulse of a positive voltage to the application of the poration pulse of a negative voltage is 50 µsec or less.
