Abstract:
A method for preparing amantadine, comprising the following steps: (1) under the catalysis of a Lewis acid, performing a chlorination reaction on adamantane and chlorine in a solvent, and removing the solvent and a catalyst-containing residue in the reaction solution, so as to obtain the chlorination reaction product; and (2) mixing the chlorination reaction product with urea, and performing an amination reaction, so as to obtain the amantadine. Cheap chlorine is used for preparing amantadine, having a low preparation cost and high product purity, and avoiding the problem of difficult treatment of three wastes in a bromination reaction; after a chlorination reaction is completed, only a solvent and a catalyst-containing residue are removed to perform a next reaction without purifying a product, having simple steps and a small amount of generated three wastes; and unreacted adamantane can be reused, further reducing three wastes, and facilitating industrialization. Results of the examples show that the purity of the prepared amantadine can reach 99.5% or more.
