Abstract:
Disclosed is a method for preparing (S)-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid by means of a chemoenzymatic method, comprising: using a racemic 1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid as a substrate, stereoselectively catalyzing the R-isomer with a D-amino acid oxidase and generating a corresponding imidic acid by means of oxidative dehydrogenation, wherein the S-isomer is not catalyzed and remains in the reaction system, the imidic acid is made into a racemic substrate under the action of an imidic acid reducing agent, and the R-isomer therein is stereoselectively catalyzed under the action of the D-amino acid oxidase, and thus the S-isomer is prepared. The present method has a reaction yield that can reach 80.6% or more, an ee value of > 99%, and has characteristics such as mild reaction conditions, a strong stereoselectivity, a high reaction efficiency, a high yield, and relatively simple processes.
