Abstract:
A method for purifying and concentrating DNA in forensic samples using a selective filtration column, specifically as follows: adding a cell fluid having undergone digestion and pyrolysis into an inner pipe of a selective filtration column, and at the same time, centrifuging the selective filtration column in a centrifuge; adding 5-100 microliters of water into the selective filtration column, oscillating for 30 seconds, and then transferring the aqueous solution in the selective filtration column to a clean centrifuge tube, and at such time, the solution in the centrifuge tube is a purified and concentrated solution. After using a lysis buffer to perform pyrolysis and digestion on residual human cells in a sample, DNA is then completely free in a solution. Solid particles in the sample are centrifuged away by means of a centrifuge tube, and then by means of an ultrafiltration membrane at a bottom portion of a selective filtration column, said ultrafiltration membrane being permeable to small molecule substances and impermeable to the macromolecule substance that is DNA under a condition of high-speed centrifuging, purification of the macromolecule substance DNA is made possible.
