Abstract:
Provided are a transcriptional regulation factor specifically responsive to D-2-hydroxyglutaric acid and an application thereof in biological detection of D-2-hydroxyglutaric acid. The transcriptional regulation factor is named as DhdR, the nucleotide sequence of the factor is shown as SEQ ID NO. 1, and a D-2-hydroxyglutaric acid biosensor BD2HG-0 and a D-2-hydroxyglutaric acid biosensor BD2HG-1 constructed by using the transcriptional regulation factor can be used to perform detection on biological samples containing D-2-hydroxyglutaric acid. Experiments show that the biosensors can realize detection having high sensitivity, specificity and accuracy on D-2-hydroxyglutaric acid, and can be used for the detection of D-2-hydroxyglutaric acid, diagnosis and treatment of D-2-hydroxyglutaric acid related diseases, and development of targeted drugs.
