Abstract:
A two-photon microscopic imaging system having low light damage. An excitation light emitted by a femtosecond pulse laser (1) is modulated by a photoelectric modulator (2), and can then realize, by means of a first movable reflection mirror (4) and a second movable reflection mirror (11), switching between a first scanning module (8) and a second scanning module (10). The first scanning module (8) can perform resting-state imaging of biological living tissue by means of raster scanning, and the second scanning module (10) can realize video-level fast imaging, such that fast-changing neural activity in the biological living tissue can be observed. A photoelectric modulator is controlled in real time during an imaging process, such that gating excitation at a specific pixel point, such as a target cell, can be realized, so as to reduce ineffective radiation of laser light on an object under test, thereby decreasing light damage. The two-photon imaging system can reduce the thermal damage effect of traditional two-photon excitation, and is suitable for long-term living body imaging.
