Abstract:
PURPOSE:To prepare a high purity heparitinase useful for the study of heparitin sulfuric acid, easily in large quantities, by the column chromatographic treatment of a prepiminary purified heparitinase solution using an adsorbent obtained by treating a heparin-immobilized agarose with heparin. CONSTITUTION:Bacterial cells prepared by culturing heparitinase-producing bacteria, e.g. Flavobacterium heparinum in a medium containing heparin, are disintegrated by ultrosonic treatment, etc. The disintegrated cells are extracted, and preliminary purified by protamine sulfate treatment, hydroxyapatite fractionation, etc. The purified heparitinase free from other acidic mucopolysaccharase is obtained by the column chromatography of the preliminary purified heparitinase using a heparin- immobilized agarose treated with heparin as an adsorbent.
