1462686 Steroid 9,11-epoxides OMNI RESEARCH Inc 13 Feb 1974 [21 Feb 1973] 53278/75 Divided out of 1462685 Heading C2U The invention comprises compounds I, VIIa, VIIb and 14 infra wherein "Ac" is acyl except in compound I where it is acetyl only, and their preparation from compounds IIIa and IIIb infra (see also Specification 1,462,685) by the routes shown or variants thereof described below Remarks (i) in III#IV, MeMgI may replace MeMgBr, and IV is preferably not isolated; (ii) in IV#V acetic anhydride or propionic, butyric, valeric, heptanoic or cyclopentylpropionic chloride or anhydride may replace acetyl chloride; (iii) in V#VI other peracids may be used; (iv) V and VI are each obtained as mixtures of the 20-epimers which may be separated by fractional crystallization and chromatography; (v) VI#VIIa#VIIb is conventional; (vi) VIIa#5 and VIIb#11 uses Br 2 +HBr; (vii) 11#5 uses HCl/MeOH; (vii) 5#1 uses potassium acetate; (viii) compound 14 is initially obtained as its 20-ethylene acetal and is then hydrolysed. An alternative route from IV to VII involves quenching IV with aq. NH 4 OH to give 3#- hydroxy - 9#,11# - epoxy - 16α - methyl - 5α- pregnan - 20 - one and/or a 3 - acylate thereof, which after full 3-acylation or 3-saponification is reacted with O 2 /KOBut to give a 17-hydroperoxide reduction of which with Zn/AcOH yields VIIa or VIIb.
