Abstract:
PROBLEM TO BE SOLVED: To obtain a compound having squalene synthase inhibitory action, hypocholesterolemic action, triglyceride-depressive action, thus useful for preventing and treating hyperlipemia and arteriosclerosis, etc.
SOLUTION: This compound (salt) is expressed by formula I [R is an (OH- substituted) lower alkyl; X is a (substituted) carbamoyl or (substituted) heterocycle bearing H atom capable of deprotonation; R1 is a lower alkyl; W is a halogen), e.g. (3R,5S)-N-methanesulfonyl-7-chloro-5-(2,3- dimethoxyphenyl)-1-(3-hydroxy-2,2-dimethylpropyl)-2-oxo-1,2,3,5-tetrahydro-4,1- benzoxazepine-3-acetamide. This compound of formula I is obtained by condensation reaction between a compound of formula H and a compound of formula III in a solvent (e.g. DMF) in the presence, if necessary, of a base (e.g. triethylamine). An orally administrative agent, in the form of pharmaceutical preparation, contains 0.01-99wt.% of the compound of formula I.
