PURPOSE: To obtain a carrier supporting an immobilized enzyme in high stability of enzymatic activity, formable to arbitrary form by simple operation and utilizable as an enzymatic sensor, etc., by dissolving an enzyme solution in a β-chitin dispersion and forming the solution to immobilize the enzyme.
CONSTITUTION: Gladius of sagittated calamary is pulverized with a feather mill, put into 1N aqueous solution of sodium hydroxide, heated at 90°C for 1hr, washed with water, immersed in 0.1N HC1 solution at room temperature for 1hr, washed with water, heated in 1N aqueous solution of sodium hydroxide at 90°C for 1 hr, washed with water and dried in an oven at 50°C for 5 hr to obtain β-chitin. The β-chitin is crushed, water is added to the dried powder to a concentration of 0.2wt.% and the mixture is vigorously agitated to increase the viscosity and form a pasty gel. An aqueous solution of an enzyme (e.g. β-D-glucose oxidase) is added to the gel and the mixture is formed in the form of a sheet by a water flow-down paper-making process and dried in air at 4°C for 15hr to obtain the objective carrier having an enzyme immobilized to β-chitin.
