Abstract:

To obtain the subject new gene and its 3'-terminal untranslated region, comprising a coat protein gene of a watermelon mosaic virus and its 3'-terminal untranslated region having a specified sequence and useful for the production, etc., of plants resistant to the watermelon mosaic virus.

The new coat protein gene of a watermelon mosaic virus and its 3'-terminal untranslated region have a base sequence represented by the formula and are useful for the production, etc., of a plant resistant to the watermelon mosaic virus. This gene is obtained by separating the watermelon mosaic virus (KY strain) from a cucumber, extracting an RNA thereof by a conventional method, carrying out the reverse transcription-polymerase chain reaction (RT-PCR) of the extracted RNA with two kinds of synthetic primers composed of a part of the 5'-terminal and 3'-terminal of the coat protein gene of the watermelon mosaic virus, integrating the resultant cDNA of the coat protein gene of the watermelon mosaic virus into a vector, then transforming Escherichia coli, cloning the transformant and recovering the DNA from the selected clone.
