Abstract:

To provide a common method applicable to all variants generally used as a DNA marker for genetic analysis.

The analysis method comprises preparing a polynucleotide target involving nucleotide of a specified position and an oligonucleotide probe fixed on a support which probe complementary to the target and terminates at the specified position or its vicinity, and performing steps of (a) incubating the target with the probe to form a duplex chain, (b) incubating the duplex chain with a labelled oligonucleotide complementary to the target under a ligation condition and (c) monitoring the ligation of (b) as one exhibiting point mutation at the specific position in the target.
