Abstract:

To efficiently identify the sequence of a target DNA chain by terminating synthesis of a DNA chain with a substrate not containing adenine, cytosine, guanine and thymine, labeling the DNA chain and irradiating the DNA chain with light rays to detect a labeled DNA chain.

Synthesis of a DNA chain is terminated with a substrate not containing adenine, cytosine, guanine and thymine and the DNA chain is labeled by a fluorescence-label, etc. The labeled DNA chain is applied to plural parts in a gel electrophoresis so as to pass the gel electrophoresis and to subject to electrophoresis in plural channels. An electric potential is established in the whole electrophoresis slab. DNA samples are separated to a fluorescence labeled DNA bands according to the sizes of DNA fragments in the electrophoresis slab. The DNA chain is photoelectrically irradiated with light rays by a laser diode and a sensor and the light at the release wavelength of the labeled DNA is detected to efficiently identify the sequence of the target DNA chain.
