Abstract:

To provide a method for nonspecifically amplifying a nucleic acid regardless of the base sequence of the nucleic acid, and to provide a reagent therefor.

The method for amplifying the nucleic acid comprises the following steps: (1) a step for synthesizing a DNA fragment having a common sequence 1 not possessed by the nucleic acid at the 5' terminus; (2) a step for adding a homopolymer sequence to the 3' terminus of the DNA fragment; (3) a step for synthesizing a DNA fragment having common sequences at both 5' and 3' terminuses by using a primer having a common sequence 2 not possessed by the nucleic acid at 5' terminus and complementary to the homopolymer sequence at the 3' terminus, and a DNA polymerase; and (4) a step for carrying out a polymerase chain reaction by using a primer 1 having at least the partially same base sequence as the common sequence 1, and a primer 2 having at least the partially same base sequence as the common sequence 2.
