Abstract:

NEW MATERIAL: A compound of formula I (B is a purine, 7-deazapurine, or pyrimidine moiety covalently bonded to the C1'-position of the sugar moiety, provided that when B is purine or 7-deazapurine, it is attached at the N9-position and when B is pyrimidine, it is attached at the N1-position; A is formula II, formula III, etc.; the dotted line is a linkage joining B and A, which is attached to the 8-position of the purine, 7-position of the 7-deazapurine or the 5-position of the pyrimidine; x, y and z are each H, OH, formula IV, etc.).

USE: Diagnostic agents for genetic disorders, infections, etc.

PROCESS: For example, a nucleotide of formula V is dissolved in 0.1 M acetate buffer (pH 6.0). The solution is reacted with a mercury salt such as mercuric acetate to give an Hg-nucleotide of formula VI, which is then reacted with an allylamine in the presence of K2PdCl4. The resultant product is reacted with a compound of the formula, MA (M is a functional group) to give the modified nucleotide of formula I.
