PURPOSE: The titled cellular surface antigen recognition type antibody, produced by a hybridoma prepared by using a cellular strain (MKN-45) derived from a gastric low differentiation type gladular cancer as an immunization source, having a light chain of IgG1(K) and capable of exhibiting positive reaction to cancer cells, e.g. specific gastric cancer or esophageal cancer.
CONSTITUTION: A monoclonal antibody obtained by fractionating a cell extract of an established cell (MKN-45) derived from a gastric low differentiation type glandular cancer, mixing the resultant high-molecular fraction having 700,000W1,500,000mol.wt. with complete Freund's adjuvant, immunizing an animal, e.g. mouse, with the resultant mixture, collecting a cell producing an antibody, carrying out cell fusion with a myelomatous cell of an animal of the same species, cloning the resultant cell to give a cloned hybridoma, transplanting the resultant cloned hybridoma to the abdominal cavity of an animal, e.g. BALB/C mouse, to produce the aimed antibody capable of exhibiting positive reaction to cancer cells of gastric cancer (MKN-45 or KATO-III), esophageal cancer (TE-3), lung cancer (PC-10) and colonic cancer (Colo201) and recovering and purifying the antibody from the ascites.
