Abstract:
To provide mutated immunoglobulin-binding domains of Protein A useful in the field of affinity chromatography, more particularly, useful in the affinity chromatography of immunoglobulins, as well as multimers of mutated domains and separation matrix comprising the mutated domains or multimers thereof.SOLUTION: Disclosed is an Fc binding polypeptide having improved alkali stability comprising a mutant of Fc binding domain of Staphylococcal Protein A (SpA), as defined by one of SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO:3, SEQ ID NO: 4, SEQ ID NO: 5, SEQ ID NO:6, SEQ ID NO:7, SEQ ID NO:8, SEQ ID NO 26 or SEQ ID NO 27, where at least the alanine residue at position 42 in SEQ ID NOs: 4-7 has been mutated to arginine and/or at least the asparagine residue at position 37 of SEQ ID NOs: 4-7 has been mutated to glutamic acid.SELECTED DRAWING: Figure 1
