PURPOSE: To provide a new compound active on Gram-positive bacteria, highly sustainable in its antibacterial activity in plasma, thus useful as an antibacterial agent.
CONSTITUTION: The compound of formula I [R1 is O, OH plus H, or group of OCOR6 (R6 is 1-3C straight chain alkyl) plus H; R2 and R3 are each H or group of COR6; R4 is 1-4C straight chain alkyl; R5 is (substituted) 1-10C chain alkyl or aralkyl] or a salt thereof. The compound of the formula I can be obtained by reaction of a compound of formula II with a 2-deoxy-L-saccharide of formula III (R10 is CH2CH3) in the presence of silver perchlorate. The compound of the formula II, which is also a new compound, can be obtained, for example, by reaction of an peroxide with a compound of formula IV produced from mydecamycin A3 followed by acid hydrolysis of the product. And, the compound of the formula III can be obtained, for example, by introducing alkyl side chain into a β-glycoside of formula V produced from erythromycin followed by acid hydrolysis and then reaction of the product with aldolithiol-2(2,2'-dipyridyl disulfide) and tri(n-butyl)phosphine.
