PURPOSE: To allow convenient and accurate immunological measurement of various vitamin K dependent plasma proteins (PIVKA) in an organic sample using an anti-PIVKA-II monoclonal antibody.
CONSTITUTION: 2-antibody sandwich method is preferably employed in the measurement of various PIVKA wherein a solid phase anti-PIVKA-II monoclonal antibody is used as a first antibody and an antibody for the normal protein in each PIVKA to be measured is used as a label for second antibody. In case of measurement of PIVKA-VII (IV, V), an anti-VII-th (IV-th, V-th) factor antibody for the common antigen of PIVKA-VII (IV, V) and factors thereof is used and in case of measurement of PIVKA-C (S, Z), an anti-protein C (S, Z) antibody for the common antigen of the PIVKA-C (S, Z) and protein C (S, Z) is used. Conventional carrier, e.g. microtiter plate or red blood cell, can be used as the solid phase carrier of the first antibody and a determinable substance, e.g. a fluorescent substance or a pigment, can be employed as a labeling substance for the second antibody.
