Abstract:
NEW MATERIAL:A neutral saccharide-containing luteinizing hormone which contains more than 30% of neutral saccharide chain components in the saccharide chain component composition of the beta-subunit. USE:kidney growth promoter. PREPARATION:For example, swine hypophysis is homogenized together with 0.15M aqueous ammonium sulfate and phenylmethylsulfonyl fluoride, adjusted in its pH to 4.0, centrifuged to collect the supernatant. Then, the pH of the supernatant is adjusted to 6.5-7.0, and ammonium sulfate is added to the supernatant to form precipitate. The precipitate is collected and suspended in a buffer, transferred into a dialysis tube to effect dialysis, then freeze-dried. The dried powder is dissolved in a carbonate buffer solution and fractionated by passing through a molecular sieve column to collect the fractions of about 40,000 molecular weight. The fraction is further fractionated using a Con A- immobilized column, the adsorbate is eluted and purified by the isoelectric electrophoresis to obtain the subject kidney growth promoting factor.
