Abstract:
Aminoheterocycles of formula (I) and their acid addition salts are new (n is 1-3, A is nitrogen or CH. X is methylene, or provided A is not CH, also methyleneoxy. When A is CH, R is phenyl opt. 2-fluoro substd. and when A is N, R is phenyl (opt. substd. by halo, nitro, 1-4C alkyl or alkoxy, or trifluoromethyl), pyridyl or cyclohexyl. NR1R2 is amino or methylamino (but not when X is CH2O or when A is N, X is CH2 and n is 1), di(1-4C) alkylamino, pyrrolidino, piperidino, morpholino or 4-methylpiperazino. When A is CH, R3 is H and when A is N, R3 is H, halo, 1-4C alkyl or alkoxy or represents two methoxy gps.). (I) have CNS, esp. analgesic and antidepressant activities. Cpds. tested had acute intraperitoneal LD50 in mice 35-170 mg. per kg. and analgesic ED50 (phenylbenzoquinone test) of 1-30mg. per kg. About 60 cpds. e.g. where A is N or CH and X is CH2 or where A is N and X is CH2O; R1R2N is dimethylamino; n is 1 or 2; R is phenyl and R3 is H are specifically claimed.
