Abstract:
PURPOSE: To obtain the subject compound used for betamethasone source material, etc., by dehydrating a 17α-hydroxy-17β-cyanoandrosta-diene-3-one to an novel intermediate, bringing a CN group to a methyl ketone and methylating after epoxidizing.
CONSTITUTION: The 16 β-methyl-steroid expressed by formula IV is obtained by treating a 17α-hydrxy-17β-cyanoandrosta-5,9(11)-diene-3-one expressed by formula I (K is oxo, ketal, etc.), with a dehydrating agent to prepare a new intermediate expressed by formula III, when K of the compound is oxo, bringing the compound to a compound modified by a suitable blocking agent of the oxo-residue, acting an organometallic methylation agent and hydrolyzing an amine intermediate to bring to a methyl ketone compound which is a novel intermediate expressed by formula IV, treating the compound of formula IV with an epoxidizing agent in a basic medium to bring to a novel intermediate expressed by formula V, successively protecting the 20-keto functional group, treating the protected compound with the organometallic methylation agent and removing the protecting group.
