PURPOSE: To obtain a piperidine derivative expressed by a specific formula and useful as an agent for the treatment and prevention of various senile dementia such as senile dementia of Alzheimer type and as an anticholinesterase agent.
CONSTITUTION: The compound is expressed by formula I [R1 is a univalent group derived from a (substituted) pyridine; X is (CH2)m ((m) is 0-7), O(CH2)n, S(CH2)n, NH(CH2)n, SO2NH(CH2)n, etc.; the ring A is group of formula II; R2 is a lower alkyl, a (substituted) benzyl, a (substituted) benzoyl, pyridine, 2- hydroxyethyl, pyridylmethyl, etc.], e.g. N-[4'-(1'-benzylpiperidine)ethyl]-4- isonicotinamide hydrochloride. The compound can be produced e.g. by reacting an acid halide of formula III (Hal is a halogen) with a piperidine derivative of formula IV in an organic solvent (e.g. chloroform) in the presence of a dehydrochlorination agent (e.g. sodium carbonate) at room temperature, under cooling with water or under heating.
