Abstract:
PURPOSE: To obtain a novel intermediate for a phenyl-substituted prostaglandin I2 which is useful as a variety of medicines.
CONSTITUTION: A compound of formula I [Z1 is H, a protecting group for OH; X1 is H, halogen, OH, SH, amino; X2 is a halogen,-Y1h(CR1R2)mAn(CH2)pY2(CR3R4)rQs (Y1, Y2 are O, S; A is a divalent group ; R1-R4 are H, a 1-4C alkyl, a 1-4C alkoxy; Q is H,-COOH, cyano; h, n, q, s are 0, 1; m, p, r are 0-5]. The compound of formula I is obtained by rearrangement of a furan derivative of formula II in the presence of an acid catalyst, isomerization under acidic or basic conditions and, when needed, protection of the hydroxyl group. Further, the reaction of a compound of formula I with a nucleophilic reagent gives a novel phenyl-substituted hydroxycyclopentanones, the reaction of the product with a reducing agent affords a novel phenyl-substituted hydroxycyclopentanes, and cyclization forms a phenyl-substituted prostaglandin I2.
