Abstract:

PROBLEM TO BE SOLVED: To obtain the subject antibody not reacting with hemoglobin (HbAo) and useful for diagnosis, etc., of diabetes mellitus by injecting a glycosylated hemoglobin component as an antigen into an organism having antibody-forming ability, immunizing the organism and acquiring antibody from the organism.
SOLUTION: This polyclonal antibody is obtained by injecting a compound represented by formula I {(m) is (l) to (q) [(q) is a numerical value corresponding to 25% of weight of carrier protein binding to a hapten spacer group]; T is a carrier protein; A is a spacer of formula II [X is S or NH; (n) is 1-4; B is an organic group containing succinimide group] and having chain length with 10-20 number of atoms} as an antigen into an organism having ability to form an antibody to immunize the organism, then collecting antiserum from the organism and recovering the antibody from the antiserum. The polyclonal antibody exhibits high affinity for glycosylated hemoglobin (HbA1c) and does not bind the hemoglobin (HbAo) and is useful for diagnosis, etc., of diabetes mellitus as use for determining HbA1c which becomes a parameter for controlling glucose metabolism.
