Abstract:

PURPOSE: To provide a method for preparing the subject outer membrane protein usable as bacteriocidal antibody inducer by which a lipooligosaccharide is depleted by extracting the outer membrane without the inner membrane from the whole membrane of a Gram-negative cocci with a specific nonionic surfactant, and extraction-treating the outer membrane with an amphoteric surfactant.
CONSTITUTION: Gram-negative cocci (e.g. Neisseria menigitidis) are freeze-dried to be extinct, suspended in a hypotonic solution, and subjected to a centrifugal separation to separate a cytoplasm extract as a supernatant and to gather the whole membrane as a pellet. The whole membrane of the cocci is subjected to an extraction treatment with a 1% solution of a polyoxyethylene surfactant to remove the inner membrane and to prepare the outer membrane of the cocci. The obtained outer membrane is subjected to the extraction treatment with a buffer solution containing 1% amphoteric ionic betain surfactant of the formula [R1 is a 10-16C alkyl: R2 is carboxyl or sulfonyl; (n) is ≥1, preferably 2 or 3] to remove lipooligosaccharide and to provide the objective outer membrane protein effective for treatment or the like of bacterial meningitis or the like and capable of inducing bacteriocidal antibody.
