Abstract:
To provide a method for producing 2'-O-alkylated guanosine 3'-O- phosphoroamidite from 2, 6-diamino-9-purine.
This method for producing 2'-O-alkylated guanosine 3'-O- phosphoroamidite comprises the steps of producing 2,6-diamino-9-(2'-O-alkylated ribofuranosyl)purine by alkylating 2, 6-diamino-9-(ribofuranosyl)purine, producing 2'-O-alkylated guanosine by deaminating the 2,6-diamino-9-(2'-O-alkylated ribofuranosyl)purine, protecting 5'-hydroxyl part of the 2'-O-alkylated guanosine and forming a phosphite of 3'-position of the 5'-protected 2'-O-alkylated guanosine to provide the objective 2'-O-alkylated guanosine 3'-O- phosphoroamidite.
