Abstract:
PURPOSE: To efficiently obtain the subject enzyme with high transglucocylation activity, useful for producing cyclodextrins to be use in the fields of foods, medicines or cosmetics by culturing Bacillus DK-1139 strain (mutant) and collecting the cultured product.
CONSTITUTION: A medium is inoculated with DK-1139 strain (FERM P-13119) belonging to Bacillus or mutant thereof to conduct culture at 30-40°C at pH 9-11 for 1-7 days, and the cultured fluid is either centrifuged or filtered to separate microbes. The supernatant or filtrate is subjected to absorption chromatography, i.e., passed through a β-cyclodextrin polymer column and adsorbed thereto; the adsorbate is then eluted with distilled water and put to gel filtration to recover a single active fraction, thus obtaining the objective enzyme with such high transglucosylation activity as to hydrolyze mainly starch or starch-based glucid into α-β-γ-cyclodextrins.
