Abstract:

PURPOSE: To obtain in an industrially advantageous way neotrehalose, a non- reducing disaccharide having good-quality, profound sweetness, serving as an energy source, thus useful as a sweetener by making α-amylase act on a starch substance.
CONSTITUTION: A starch substance (e.g. dextrin) is dissolved in hot water and the pH level of the resulting solution is adjusted to 5.6; 100 units per g of the starch substance, of a saccharifying α-amylase is added to the solution at 60°C followed by reaction for 20hr and then heating to deactivate the enzyme. The resulting reaction liquor containing neotrehalose formed is conventionally decolored using active carbon. Then, the resultant liquor is put to a column chromatography using a salttype strongly acidic cation exchange resin to collect a fraction of neotrehalose content, which is, in turn, concentrated to crystallize and collect the neotrehalose, thus industrially obtaining the objective neotrehalose highly utilizable metabolically and having the above-mentioned advantages.
