PURPOSE: To produce a novel DNA useful for obtaining a composition having Factor VIII:C activity without accompanying propagation of virus disease.

CONSTITUTION: This DNA composition contains (A) a first manifestation cassette containing the first gene coding the first peptide having substantially same sequence as the first signal sequence and an A domain of a human VIII:C factor under the rule of transcription and a translation controlling signal operating in a host cell and (B) a second manifestation cassette containing the second gene coding a peptide containing having substantially same as the second signal sequence and a C domain of a human VIII:C factor but not containing B domain under the rule of transcription and a translation controlling signal operating in the host cell. The DNA composition is obtained by arranging so as to the manifestation cassettes A and B to be simultaneously manifested in a eucaryotic phenotypic transformation host cell in a trans form.
