Abstract:
PROBLEM TO BE SOLVED: To obtain a new isolated DNA coding for Sca I restriction endonuclease derived from Streptomyces caespitosus and capable of giving an enzyme for preparing fragments for molecular cloning and gene characterization.
SOLUTION: This new isolated DNA codes for Sca I restriction endonuclease derived from Streptomyces caespitosus and is useful for cutting a DNA molecule into exact fragments for molecular cloning and gene characterization. This isolated DNA is obtained by the following process: a genome DNA is extracted from Streptomyces caespitosus by conventional means, and using the genome DNA, a plasmid library is made up and a Sea I restriction endonuclease gene is cloned by e.g. endo-blue method.
