Abstract:
PURPOSE: To readily and inexpensively obtain a highly purified sphingo glycolipid for a raw material of a medicine and a reagent, etc., by acting glycosidase to an oligosaccharide higher than a disaccharide or a glycoside and a ceramide and a sugar residue is bonded with the ceramide by a sugar- transfer reaction.
CONSTITUTION: An oligosaccharide higher than a disaccharide (e.g. cellobiose) or a glycoside and a ceramide are suspended in a 0.1M acetic acid buffer solution (pH 5.0) containing taurodeoxycholic acid, a glycosidase (e.g. β-glycosidase) is added to the suspension and reacted for 2hr to bind a sugar residue with the ceramide by a sugar-transfer reaction to generate the objective sphingo glycolipid, the reacted solution is subjected to ODS (octadecylsilane) column chromatography and an unreacted substance, a decomposition product and a taurodeoxycholic acid, etc., are removed, and further the resultant solution is purified by subjecting to silica gel column chromatography to readily and inexpensively obtain the objective highly purified sphingo glycolipid useful for a raw material of a medicine and a reagent, etc.
