Abstract:

PURPOSE: To obtain the subject protein crystals stable and useful for the treatment, etc., of diabetics by crystallizing human insulin analogs, from a solution containing LysB28ProB29-human insulin, zinc, an organic acid and a phenolic compound in a specific pH range.

CONSTITUTION: The objective stable zinc insulin analog crystals useful for the treatments, etc., of diabetics and having relatively rapid activity-expression are obtained by diluting LysB28ProB29-human insulin to 1.8-2.5 g/l concentration with a buffer comprising acetic acid, citric acid, glycine or the like and containing at least 0.3 N organic acid, adding a phenolic compound (e.g. phenol, etc.), to the solution in approximately 0.15-0.25% based on the final volume, adjusting pH to approximately 5.9-6.2 by using ammonium hydroxide, adding the solution of a zinc salt (e.g. zinc chloride, etc.), to approximately a final concentration of 40-400 mg zinc/g LysB28ProB29-human insulin, standing at 2-8°C for ≥18 hours to precipitate crystals and centrifuging the crystals.
