PURPOSE: To obtain an L chain variable region of an antibody against a human medulloblastoma cell useful for preparation, etc., of a chimera antibody useful for treatment, diagnosis, etc., of cerebral tumor, containing plural complementarity determining regions and framework regions having a specific amino acid sequence and having low antigenicity.
CONSTITUTION: This L chain variable region(V region) of an antibody against a new cell of a human medulloblastoma containing three complementarity determining regions(CDR) comprising amino acid sequences represented by formulas I to III or a part thereof and four framework regions(FR). The L chain V region is obtained from an L chain CDR of an anti-human medulloblastoma cell monoclonal antibody ONS-M21 prepared by immunizing a mouse with the human medulloblastoma cell. A reshaped human antibody is prepared by binding an L chain V region comprising the murine L chain CDR to a human L chain C region and a human L chain FR, reshaping the L chain, binding a human H chain C region to a human H chain FR and an H chain V region comprising an H chain CDR of a murine monoclonal antibody against the human medulloblastoma cell and binding the resultant reshaped H chain to the reshaped L chain. The reshaped human antibody is useful for treatment, diagnosis, etc., of cerebral tumor.
