Abstract:

PURPOSE: To obtain a thermostable α-galactosidase having high saccharide transition activity and a complex thereof from a culture mixture by cultivating fungi belonging to the genus Candida gulliermondii.

CONSTITUTION: Candida gulliermondii H-404 (FERM P-11026) is cultured in liquid medium usually comprising melibiose, etc., or a decomposed product thereof alone or sucrose, powdered wheat gluten, lactose or starch and melibiose as an inducer, as a carbon source and the following nitrogen source. Namely, the nitrogen source is an organic or inorganic nitrogen source such as whole fats, de-fatted soybean, corn steep liquor or yeast essence. The culture condition is 20-40°C, about 2-14 days and aerobic condition. The thermostable α- galactosidase complex having high saccharide transition activity prepared by the culture has strong saccharide transition activity of hardly receiving substrate inhibition even at about 1M substrate concentration and ≥80% transition of formed galactose.
