Abstract:

PURPOSE: To obtain the titled reagent for determination, consisting of leucylalanine and alanine dehydrogenase, having excellent quantitative property and reproducibility, usable for routine tests treating many specimens without requiring troublesome operation and useful for diagnosing pancreatic cancer, metastasis of cancer to the liver, etc.

CONSTITUTION: A reagent for determination prepared from 1W100mM leucylalanine which is a substrate specifically reactive with leucine aminopeptidase, 0.5W100 units/ml alanine dehydrogenase, 0.5W30mM nicotinamide adenine dinucleotide (NAD+), etc. The principle of the reagent in measurement is as follows. An amino acid liberated from the substrate by action of the leucine aminopeptidase is decomposed into pyruvic acid and ammonia by action of the alanine dehydrogenase and coexisting NAD+ is converted into reduced form nicotinamide adenine dinucleotide (NADH). The amount thereof converted is measured by absorbance at 340nm.
